Specifications include, but are not limited to: The purpose of this RFP is to enter into a contract to provide: services to test the percent triploidy among batches (lots) of pressure-treated larval fish for Walleye. Testing of grouped fry samples The bidder shall provide estimates of percent triploid fry for every sample and replicate sub-sample received. Estimates of timing and volume of work expected are as follows: Samples will be shipped overnight on Mondays and Wednesdays for next day processing from about April 1 through June 1. Each shipment will contain 2 – 12 bags of live fry with at least 500 individuals per bag. We anticipate 30 – 35 total bags will be shipped for testing over the course of the hatch. Each bag will represent one pressure treated lot that will be divided into 5, 100 fry samples. Each 100 fry sample will be homogenized, separated, and cellular nuclei will be isolated for analysis of triploidy by flow cytometry. Data will be analyzed and results reported to UDWR staff by the following day as percent triploid fry per 100-fry sample. Testing of individual blood samples The bidder shall provide counts of diploid and triploid individuals for every sub-sample received. Estimates of timing and volume of work expected are as follows: A total of 3 unique blood sample lots will be shipped overnight between approximately May 15 through June 10. Each lot will contain a minimum of 500 blood samples for a total of 1,500 blood samples Data will be analyzed and results reported to UDWR staff within 24 hours of submission. Lot triploidy will be reported to UDWR as number (count) of diploid and triploid individuals per 100 blood samples within each lot (e.g., 5 ploidy percentages will be reported for each of the three 500-sample lots submitted; equating to total of 15 ploidy percentages across 3 lots).
