Although GCL cannot guarantee a minimum quantity of purchase, based on projections, GCL expects to sequence approximately 13 sequencing-ready libraries from a total of 1,200 fish, tagged with dual-barcoded 8 base-pair (bp) i5 and i7 Illumina Nextera DNA indexes, the estimated equivalent of up to 30,000 gigabases; 1. Sequencing platform must be compatible with Illumina Sequencing Chemistry with sequencing and P5/P7 adapter sequences already incorporated by GCL into the salmon libraries.; 2. Sequences must be collected as paired end 150bp reads (2x150bp).; 3. Sequencing platform must provide medium coverage for each library. (An average read depth of 10x across the genome with a minimum read depth of 7x within lower coverage areas of the genome.); 4. Bidder must have an in-house QC to ensure data quality as part of the synthesis process. Examples may include, but are not limited to, Bioanalyzer, TapeStation, or qPCR platforms.; 5. Resulting Sequence Data (Data) must have a minimum Quality Score of ≥ 85% of bases higher than Q30.; 6. Data must be provided to GCL as gzipped fastq files (*.fastq.gz) formatted for use in a Unix/Linux analysis pipeline and delivered via sftp download and a backup external portable hard drive.; 7. Data must be received by GCL no later than twenty (20) standard business days after bidder receives prepared libraries.
